Vector Biolabs G protein-coupled receptor kinase 5 Adenovirus (Ad-GRK5)
Heterotrimeric G protein-mediated signal transduction is a dynamically regulated process with the intensity of signal decreasing over time despite the continued presence of the agonist. This phenomenon, referred to as agonist-mediated desensitization, involves phosphorylation of the receptor by two classes of enzymes. The first is the second messenger regulated kinases such as cAMP dependent protein kinase A and protein kinase C. The second is the G protein-coupled receptor kinases (GRKs). Six members of the GRK family have been identified to date. These include rhodopsin kinase (GRK 1), two forms of beta-adrenergic receptor kinase (GRK 2 or beta-ARK 1 and GRK 3 or beta-ARK 2), IT-11 (GRK 4), GRK 5 and GRK 6. Phosphorylation of receptors by GRKs appears to be strictly dependent on the receptor being in its agonist-activated state.

StarGate cloning via the pENTRY-IBA vector provides a tool for the systematic and rapid screen of the best working expression conditions for a certain GOI, where nothing is known from literature. After initial pENTRY cloning, the GOI in the resulting Donor Vector can be easily transferred by a simple one-tube reaction into a multitude of expression vectors (Acceptor Vectors) providing different features.
IBA LifeSciences StrGate Acptr Vctr pASG-IBA103
The pASG-IBA103 vector is designed for expression of recombinant proteins with a C-terminal Twin-Strep-tag in E. coli. The vector carries an ampicillin resistance cassette for selection of transformed E. coli cells, F1 and ColE1 origin for a high plasmid copy number, and the inducible tetracycline promoter/operator for the regulated expression of proteins. The vector can be combined with any E. coli strain since the tet-promoter works independently of the genetic background of E. coli. Expression of the recombinant protein is induced by addition of anhydrotetracycline, which is a derivative of tetracycline that does not exhibit antibiotic activity. Please note that cloning into pASG-IBA Acceptor Vectors compulsorily requires the restriction enzyme Esp3I since no other MCS for the integration of a gene of interest is available. In addition to the direct cloning of the gene of interest into pASG-IBA vectors with Esp3I, another option via a so-called Entry Vector is possible.

Vector Biolabs Cre Inducible (FlexON) GCaMP6m Adenovirus (Ad-DIO-GCaMP6m)
GCaMP is a genetically encoded calcium indicator (GECI) which was created from a fusion of green fluorescent protein (GFP), calmodulin, and M13, a peptide sequence from myosin light chain kinase. GECI binds Ca2+ and induce a change in fluorescence signal, which allows measuring action potentials and other receptor activation events that trigger Ca2+ fluxes. The advantage of GECI's are that they can be genetically specified for studies in living organisms.The new novel GCaMP6 have increased ΔF/F0 and faster kinetics compared to previous GCaMP3 and GCaMP5G.This adenovirus expresses GCaMP6m in a FLEX system for Cre inducible expression.

This AAV serotype 5 virus (with Capsid from AAV5 and ITR from AAV2) expresses Cre Recombinase (tagged with nuclear localization sequence) and eGFP marker. Each gene expression is driven by its own CMV promoter. Cre recombinase is a Type I topoisomerase from P1 bacteriophage that catalyzes site-specific recombination of DNA between loxP sites. loxP is a 34 bp DNA sequence at which confers directionality. Cre recombinase is used as a tool to genetically modify genes, such as to delete a segment of DNA flanked by LoxP sites in cultured cells or experimental animals.
